چکیده

A calcium protease which degrades neurofilaments was first observed in the fan worm Myxicola infundibulum and in the squid Loligo forbesi [l] and subsequently in a mammalian system [2,3]. Many of the polypeptides in Myxicola axoplasm are degradation products of the 160 000 and 150 000 M, polypeptides as has been shown by careful peptide mapping. Moreover, there is a high probability that they result from in vivo processes since axoplasm can be extruded and solubilized in SDS in 10 s. A two hour incubation of axoplasm in the absence of calcium chelators does not alter the gel pattern [4]. sheathing followed by a 40 s sonication at 150 watts in a Braunsonic 1510 sonicator was needed to solubilize the preparation.

Developmentally regulated epitopes on a neurofilament protein visualized by monoclonal antibodies.

Two monoclonal antibodies (mabs) which recognized the 68 kDa subunit of the rat neurofilament triplet were isolated. In immunoblots with SDS-solubilized and reduced proteins, these mabs recognized their epitopes equally well in embryonic and in adult tissue. However, these epitopes were developmentally regulated in paraformaldehyde-fixed rat brain sections. They were abundant in all compartment...

Isolation and Differentiation of Neural Stem/Progenitor Cells From Subventricular Zone of One Adult Rat

Introduction: In adult mammalian brain, neural stem cells are isolated from both the dentate gyrus and subventricular zone. This study aimed to isolate neural stem cells from adult rat subventricular zone and differentiate them into neurons and astrocytes.  Methods: In this study, the whole brain was removed after full anesthesia and creating cervical dislocation. Under a microscope, subv...

P48: Protein Changes Resulted in Sub-Chronic Neurotoxicity of Bisphenol A in Rat Brain

Bisphenol A (BPA) is one of the most widely used chemicals in the plastic industry, which enter the human body through occupational and food contact. In this study, the protein changes in rat cerebral cortex was evaluate in order to evaluate the neurotoxicity of BPA. 24 adult male rats were randomly selected and divided into four groups (n=6) and each group respectively received 0, 0.5, 5 and 5...

ALTERATIONS OF ADP-RIBOSYLATION AND DNA-BREAKS IN AGING BRAIN CELLS

Neuronal and astroglial cells were prepared from whole brain of three month and 30-month- old rats for study of alterations in the nuclear poly ADP-ribosylation and DNA breaks with age. The relative purity of the cell preparations was confirmed by the determination of the neurofilament (low molecular weight) and glutamine synthetase content of the cells using ELISA. An increase (75%) in th...

High neuronal/astroglial differentiation plasticity of adult rat hippocampal neural stem/progenitor cells in response to the effects of embryonic and adult cerebrospinal fluids

Hippocampal neural stem/progenitor cells (hipp-NS/PCs) of the adult mammalian brain are important sources of neuronal and gial cell production. In this study, the main goal is to investigate the plasticity of these cells in neuronal/astroglial differentiations. To this end, the differentiation of the hipp-NS/PCs isolated from 3-month-old Wistar rats was investigated in response to the embryonic...
